High dietary threonine extraction by the digestive tract suggests that threonine contributes to maintain gut physiology. In the present study, we evaluated the impact of a low (6.5 g of threonine/kg diet; LT group) or a control well-balanced threonine diet (9.3 g of threonine/kg diet; C group) given to piglets for 2 weeks on ileal permeability and Na+-dependant glucose absorption capacity in Ussing chambers. The paracellular permeability was significantly increased in the ileum of LT compared to C piglets (P=.017). The Na+-dependent glucose absorption capacity showed a nonsignificant increase in the LT piglets. The aim of this study was to develop a procedure based on Gompertz function to determine the efficiency of utilization of amino acid. The procedure was applied to determine the efficiency of utilization of dietary lysine, methionine+cystine and threonine by growing pullets and based on the efficiencies were estimated the requirements for the growth phase of birds. The Gompertz function was fitted to the data of feed intake, body weight, feather-free body protein weight and feather protein weight of four strains of laying hens in the growth phase. The essential amino acids lysine and threonine are synthesized in higher plants via a pathway starting with aspartate that also leads to the formation of methionine and isoleucine. Lysine is one of most limiting amino acids in plants consumed by humans and livestock. Recent genetic, molecular, and biochemical evidence suggests that lysine synthesis and catabolism are regulated by complex mechanisms. Early kinetic studies utilizing mutants and transgenic plants that over-accumulate lysine have indicated that the major step for the regulation of lysine biosynthesis is at the enzyme dihydrodipicolinate synthase. Determination of aspartate kinase activity in maize tissues Scientia Agricola

Lysine, threonine, methionine and isoleucine are synthesized from aspartate in a branched pathway in higher plants. Aspartate kinase plays a key role in the control of the aspartate pathway. The enzyme is very sensitive to manipulation and storage and the hydroxamate assay normally used to determine aspartate kinase activity has to be altered according to the plant species and tissue to be analyzed. We have optimized the assay for the determination of aspartate kinase in maize plants callus cell cultures. Among all the assay parameters tested, the concentration of ATP/Mg and temperature were critical for enzyme activity. In the case of temperature, 35°C was shown to be the optimum temperature for aspartate kinase activity.

Distribution of soluble amino acids in maize endosperm mutants Scientia Agricola

For human nutrition the main source of vegetable proteins are cereal and legume seeds. The content of total soluble amino acids in mature endosperm of wild-type, opaque and floury maize (Zea mays L.) mutants were determined by HPLC. The total absolute concentration of soluble amino acids among the mutants varied depending on the mutant. The o11 and o13 mutants exhibited the highest average content, whereas o10, fl3 and fl1 exhibited the lowest average content. In general, the mutants exhibited similar concentrations of total soluble amino acids when compared to the wild-type lines, with the clear exception of mutants o11 and fl1, with the o11 mutant exhibiting a higher concentration of total soluble amino acids when compared to its wild-type counterpart...

Cereal seeds are poor in essential amino acids, particularly lysine, tryptophan and threonine. The amino acids lysine and threonine are synthesized in the aspartate pathway. Although most of the enzymes of the aspartate pathway have been isolated and characterized in higher plant species, the metabolism of lysine and threonine is totally unknown in sorghum. We have isolated two enzymes, aspartate kinase (AK) and homoserine dehydrogenase (HSDH) from sorghum. Optimum assay conditions were established for the determination of AK and HSDH activities. Three hundred and twenty Hy-Line W36<FONT FACE=Symbol>Ò</FONT> commercial laying hens, 39 weeks of age, were used to determine the sequence and the length of time needed for hens to recover performance characteristics after an eight-week period under graded levels of threonine deficiency. Eight experimental diets with Thr levels ranging from 0.35 to 0.53% were randomly fed with eight replicates of five hens each. La lisina (lis) y treonina (tre) son el primer y segundo aminoácidos limitantes en las dietas de cerdos para la síntesis de proteína. El objetivo del trabajo fue evaluar el efecto de diferentes niveles de lisina y treonina en dietas sorgo-pasta de soya para cerdos en crecimiento, sobre el comportamiento productivo, características de la canal y la concentración de urea en plasma. Threonine-lysine ratio on the requirements of digestible lysine in diets for broilers Animal Sciences

Two experiments were performed to determine the influence of threonine-lysine (Thr:Lys) ratio on requirements of digestible lysine (Lys) in low crude protein diets for broilers in the growth phase. In the first experiment, a total of 480 Cobb 500 male broilers were distributed in a completely randomized experimental design with four dietary Thr:Lysratio (57.0; 60.5; 64.0 and 67.5%) and with five replicates of 24 birds each. No significant differences were reported in weight gain, feed intake, poultry manure humidity, carcass and meat yields.
